Recombinant expression and purification of the antimicrobial peptide Magainin-2

Ramos, Reinaldo Rodrigues; Moreira, Susana; Rodrigues, Ana; Gama, F. M.; Domingues, Lucília

Magainin-2 (MAG2) is a polycationic antimicrobial peptide isolated from the skin of the African clawed frog Xenopus laevis. It has a wide spectrum of antimicrobial activities against Gram-positive and Gram-negative bacteria, fungi and induces osmotic lysis of protozoa. MAG2 also possesses antiviral and antitumoral properties. These activities make this peptide a promising candidate for therapeutic applications....

Wound healing activity of the human antimicrobial peptide LL37

Ramos, Reinaldo Rodrigues; Silva, João P.; Rodrigues, A. C.; Costa, Raquel; Schmitt, Fernando C.; Soares, Raquel; Guardão, Luísa; Vilanova, Manuel

Antimicrobial peptides (AMPs) are part of the innate immune system and are generally defined as cationic, amphipathic peptides, with less than 50 amino acids, including multiple arginine and lysine residues. The human cathelicidin antimicrobial peptide LL37 can be found at different concentrations in many different cells, tissues and body fluids and has a broad spectrum of antimicrobial and immunomodulatory act...

Recombinant expression and activity of cationic antimicrobial peptides

Ramos, Reinaldo Rodrigues

Tese de doutoramento em Engenharia Química e Biológica ; In the past 60 years, antibiotics have been critical in the fight against infectious disease caused by microorganisms. The increasing bacterial resistance to antibiotics is a serious public health problem. Much research has been dedicated to the development of new classes of antibiotics to overcome this situation. Antimicrobial peptides (AMPs) are genera...

Escherichia coli expression and purification of LL37 fused to a family III carb...

Ramos, Reinaldo Rodrigues; Domingues, Lucília; Gama, F. M.

The cathelicidin derived human peptide LL37 has a broad spectrum of antimicrobial and immunomodulatory activities. The large variety of biological activities makes LL37 a very promising candidate for clinical applications. The production of biologically active LL37 in large amounts with reduced costs can only be achieved using recombinant techniques. In this work, LL37 has been cloned to the N- and C-termini of...

Textile fabric depilling is an important industrial application of cellulases. The depilling effect and achievement of desirable touch properties are among the applications sought by users. This process, although effective, is associated with significant tensile strength loss. The depilling mechanism is still a subject of controversy. In this work, we introduce a new perspective in understanding of the depillin...

Novel hydrogel obtained by chitosan and dextrin-VA co-polymerization

Ramos, Reinaldo Rodrigues; Carvalho, Vera; Gama, F. M.

A novel hydrogel was obtained by reticulation of chitosan with dextrin enzymatically linked to vinyl acrylate (dextrin-VA), without cross-linking agents. The hydrogel had a solid-like behaviour with G′ (storage modulus) >> G″ (loss modulus). Glucose diffusion coefficients of 3.9 × 10−6 ± 1.3 × 10−6 cm2/s and 2.9 × 10−6 ± 0.5 × 10−6 cm2/s were obtained for different substitution degrees of the dextrin-VA (20% an...

Textile depilling : use of enzymes and cellulose binding domains

Ramos, Reinaldo Rodrigues; Pinto, João Ricardo; Sampaio, L.; Mota, M.; Gama, F. M.

Textile fabrics biopolishing is one of the most important industrial application of cellulases. These are widely used to remove fibrils and fuzz fibres from cotton fabrics, or to produce the “stone-washed” look of denim garments. The depilling effect and the achievement of desirable touch properties are among the applications sought by the users. This process, although effective, is associated to a significant ...
