Highly specific targeted drug delivery devices can be obtained with antibody-human serum albumin (mAb-HSA) conjugates. However, their conventional production involves several reaction steps including chemical modification and activation of both proteins followed by cross-linking often involving toxic chemicals. Here, we describe the enzymatic synthesis of mAb-HSA conjugates for targeted drug delivery devices us...

Bioprocessing of bamboo materials

Xingqun Zhang; Jiajia Fu; Paulo, Artur Cavaco; Chongwen Yu; Guebitz, G. M.

Bamboo culms were processed using microorganisms, and fibre bundles were obtained. Bacteria and fungi with xylanase activity were isolated from the bamboo retting system. Chemical composition analysis of the fibre bundles obtained showed that the components are mainly cellulose, hemicelluloses and lignin. An increase in cellulose and hemicelluloses content was detected along with a decrease in lignin content...

A biological approach for degumming bamboo substrates has been assessed. The ability of various commercially available enzymes, including cellulase, xylanase, pectinase and mannanase, to hydrolyze bamboo powders was investigated. In addition, a commercial cellulase preparation was applied onto bamboo fibre bundles obtained by natural retting. It was found that almost all enzymes applied can use bamboo materia...

Activated synovial macrophages play a key role in Rheumatoid Arthritis (RA). Recent studies have shown that folate receptor beta (FRβ) is specifically expressed by activated macrophages. Therefore a folate-based nanodevice would provide the possibility of delivering therapeutic agents to activated macrophages without affecting normal cells and tissues. This study shows for the first time the sonochemical prepa...

Enzymatic colouration with laccase and peroxidases : recent progress

Jiajia Fu; Nyanhongo, Gibson S.; Guebitz, G. M.; Paulo, Artur Cavaco; Kim, Suyeon

Enzymes have received significant attention as alternative catalysts to chemical auxiliaries in textile processing. For example, laccases and peroxidises are promising alternatives for bleaching and denim stone washing processes. Similarly, the ability to oxidise different phenolic substrates and dye precusors resulting in the formation of different coloured polymeric molecules is being exploited for developing...

Enzymatic hydroxylation of polypropylene (PP) was investigated in order to increase hydrophilicity. A mutant (139-3) of the P450monooxygenase from Bacillus megaterium expressed in E. coli DH5α was purified using anion exchange chromatography. Hydroxylation of PP fabrics led to a dramatic increase of hydrophilicity as indicated by a water drop dissipation time of below 1 s compared to the hydrophobic reference m...

Protein disulphide isomerase-assisted functionalization of keratin-based matrices

Fernandes, Margarida M.; Gomes, A. C.; Vasconcelos, Andreia; Munteanu, Florentina-Daniela; Tzanov, Tzanko; Gonçalves, M. Sameiro T.; End, N.

In living systems, protein disulphide isomerase (PDI, EC 5.3.4.1) regulates the formation of new disulphide bonds in proteins (oxidase activity) and catalyzes the rearrangement of non-native disulphide bonds (isomerase activity), leading proteins towards their native configuration. In this study, PDI was used to attach cysteine-containing compounds (CCCs) onto hair, to enhance compound migration within hair fib...

Changes in the bacterial community structure and diversity during bamboo retting

Fu, Jiajia; Mueller, Henry; Castro Jr., Joao V. de; Yu, Chonewen; Paulo, Artur Cavaco; Guebitz, G. M.; Nyanhongo, Gibson S.

Microbial retting is a critical step in obtaining fiber bundles from bamboo culm using indigenous microorganisms. A cultivation-independent technique for monitoring the changes in bacteria community during bamboo retting was applied in this work. This technique involves genetic profiling of PCR-amplified small-subunit rRNA and the single-strand conformation polymorphism (SSCP) gel analysis of the PCR-amplified ...

Antimicrobial and antioxidant linen via laccase-assisted grafting

Silva, Carla Manuela; Matamá, Maria Teresa; Padrão, Jorge; Kim, Suyeon; Prasetyo, E. N.; Kudanga, T.; Nyanhongo, Gibson S.; Guebitz, G. M.

A laccase from Ascomycetemyceliophthora thermophila was used to assist the binding of chitosan and catechin onto a previous enzymatically oxidized linen surface. The process consists of the pre-treatment of the linen with laccase followed by the application of chitosan in a first step and catechin plus laccase in a second step. The results presented here support the conclusion that laccase is able to oxidize ph...

Tailoring elastase inhibition with synthetic peptides

Vasconcelos, Andreia; Azoia, Nuno G.; Carvalho, A. C.; Gomes, A. C.; Guebitz, G. M.; Paulo, Artur Cavaco

Chronic wounds are the result of excessive amounts of tissue destructive proteases such as human neutrophil elastase (HNE). The high levels of this enzyme found on those types of wounds inactivate the endogenous inhibitor barrier thus, the search for new HNE inhibitors is required. This work presents two new HNE inhibitor peptides, which were synthesized based on the reactive-site loop of the Bowman–Birk inhib...
