Recombinant expression and purification of the antimicrobial peptide Magainin-2

Ramos, Reinaldo Rodrigues; Moreira, Susana; Rodrigues, Ana; Gama, F. M.; Domingues, Lucília

Magainin-2 (MAG2) is a polycationic antimicrobial peptide isolated from the skin of the African clawed frog Xenopus laevis. It has a wide spectrum of antimicrobial activities against Gram-positive and Gram-negative bacteria, fungi and induces osmotic lysis of protozoa. MAG2 also possesses antiviral and antitumoral properties. These activities make this peptide a promising candidate for therapeutic applications....

Biocompatibility of poly(lactic acid) with incorporated graphene-based materials

Pinto, Artur Moreira; Moreira, Susana; Gonçalves, Inês; Gama, F. M.; Mendes, Adélio; Magalhães, Fernão D.

The incorporation of graphene-based materials has been shown to improve mechanical properties of poly(lactic acid) (PLA). In this work, PLA films and composite PLA films incorporating two graphene-based materials – graphene oxide (GO) and graphene nanoplatelets (GNP) – were prepared and characterized regarding not only biocompatibility, but also surface topography, chemistry and wettability. The presence of bot...

Experimental research on masonry wall and timber elements connection

Moreira, Susana; Oliveira, Daniel V.; Ramos, Luís F.; Lourenço, Paulo B.

Studies developed for the past years and recent earthquakes have brought to light the importance that efficient connections between structural elements can have in the global behaviour of the structure. Although out-of-plane mechanisms of masonry walls are considered to be local, the reality is that their occurrence can lead to total collapse of the structure. The scarcity of data, both at experimental and nume...

Physical-chemical properties of cross-linked chitosan electrospun fiber mats

Sencadas, V.; Correia, D. M.; Ribeiro, C.; Moreira, Susana; Botelho, Gabriela; Gómez Ribelles, J. L.; Lanceros-Méndez, S.

Chitosan fiber mats were successfully processed by electrospinning. The as-spun fiber mats were neutralized with ethanol and cross-linked with glutaraldehyde. A decrease of the fiber average diameter from 243 ± 43 nm down to 215.53 nm was observed for the neutralized and cross-linking chitosan membrane. It was found that the processing conditions do not alter the initial deacetylation degree of the polymer. Pol...

Bacterial cellulose : long-term biocompatibility studies

Pértile, Renata Aparecida Nedel; Moreira, Susana; Costa, Rui M. Gil da; Correia, Alexandra; Guardão, Luísa; Gartner, Fátima; Vilanova, Manuel

The bacterial cellulose (BC) secreted by G. xylinus is a network of pure cellulose nanofibers, which has high crystallinity, wettability and mechanical strength. These characteristics make BC an excellent material for tissue engineering constructs, noteworthy for artificial vascular grafts. In this work, the in vivo biocompatibility of BC membranes produced by two G. xylinus strains was analyzed through histolo...

Bacterial cellulose modified using recombinant proteins to improve neuronal and...

Pértile, Renata Aparecida Nedel; Moreira, Susana; Andrade, Fábia K.; Domingues, Lucília; Gama, F. M.

A wide variety of biomaterials and bioactive molecules have been applied as scaffolds in neuronal tissue engineering. However, creating devices that enhance the regeneration of nervous system injuries is still a challenge, due the difficulty in providing an appropriate environment for cell growth and differentiation and active stimulation of nerve regeneration. In recent years, bacterial cellulose (BC) has em...

Recycling of cellulases in lignocellulosic hydrolysates using alkaline elution

Rodrigues, A. C.; Leitão, Alexandre F.; Moreira, Susana; Felby, Claus; Gama, F. M.

The recovery of cellulases from lignin, lignocellulosic hydrolysates and cellulose by alkaline washes at pH 9 and 10 was examined. The effect of the pH on the structural stability of purified Cel7A was analyzed by circular dichroism. Purified Cel7A showed conformational changes at pH 9 and 10 that were reversible at pH 4.8. Temperature influenced the enzymatic hydrolysis of wheat straw and may be critical for t...

Experimental study on the seismic behavior of masonry wall-to-floor connections

Moreira, Susana; Oliveira, Daniel V.; Ramos, Luís F.; Fernandes, Raquel; Guerreiro, João; Lourenço, Paulo B.

The global structural performance of masonry buildings, under earthquake loading, is affected by the efficiency of wall-to-floor connections, since they assure the continuity of the energy path and prevent the occurrence of most of the local collapse mechanisms. In fact, out-of-plane behaviour of masonry walls observed in recent seismic events showed the critical importance of proper connections in historical b...

Inhibition of IL-10 production by maternal antibodies against group b streptoco...

Madureira, Pedro; Andrade, Elva Bonifácio; Gama, Bernardo; Oliveira, Liliana; Moreira, Susana; Ribeiro, Adília; Neves, Margarida Correia

Group B Streptococcus (GBS) is the leading cause of neonatal pneumonia, septicemia, and meningitis. We have previously shown that in adult mice GBS glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is an extracellular virulence factor that induces production of the immunosuppressive cytokine interleukin-10 (IL-10) by the host early upon bacterial infection. Here, we investigate whether immunity...
