Tuberculosis associates with a wide spectrum of disease outcomes. The Beijing (Bj) lineage of Mycobacterium tuberculosis (Mtb) is suggested to be more virulent than other Mtb lineages and prone to elicit non-protective immune responses. However, highly heterogeneous immune responses were reported upon infection of innate immune cells with Bj strains or stimulation with their glycolipids. Using both in vitro and...

The authors are grateful to Drs. Manuel Teixeira da Silva, Fernando Rodrigues, Margarida Correia-Neves and Paul S. Redford for critically reading this manuscript and thank the personnel at the ICVS animal house facility for excellent animal husbandry. ; CD4+ Th1 cells producing IFN-γ are of extreme importance in controlling infections by Mycobacterium tuberculosis both in mice and in men. In addition to IFN-γ-...

Almeida, A. J.; Matute, D. R.; Carmona, J. A.; Martins, M.; Torres, I.; McEwen, J. G.; Restrepo, A.; Leão, Cecília; Ludovico, Paula

The aim of this study was to evaluate genome size and ploidy of the dimorphic pathogenic fungus Paracoccidioides brasiliensis. The cell cycle analysis of 10 P. brasiliensis isolates by flow cytometry (FCM) revealed a genome size ranging from 26.3+/-0.1Mb (26.9+/-0.1fg) to 35.5+/-0.2Mb (36.3+/-0.2fg) per uninucleated yeast cell. The DNA content of conidia from P. brasiliensis ATCC 60855-30.2+/-0.8Mb (30.9+/-0.8f...

New insights into the cell cycle profile of Paracoccidioides brasiliensis

Almeida, A. J.; Martins, M.; Carmona, J. A.; Cano, L. E.; Restrepo, A.; Leão, Cecília; Rodrigues, Fernando José dos Santos

The present work focuses on the analysis of cell cycle progression of Paracoccidioides brasiliensis yeast cells under diVerent environmental conditions. We optimized a Xow cytometric technique for cell cycle proWle analysis based on high resolution measurements of nuclear DNA. Exponentially growing cells in poor-deWned or rich-complex nutritional environments showed an increased percentage of daughter cells in ...
